Inmunoterapia adoptiva con citoquinas y linfocitos activados en un modelo experimental de tumor neuroectodérmico primitivo

Resumen

Se ha realizado un estudio experimental mediante protocolos de inmunoterapia adoptiva sobre un tumor neuroectodérmico primitivo de origen murino con capacidad de diferenciación neuroblástica (tumor IL-7). Los resultados obtenidos muestran que la administración intratumoral de interferón-alfa 2b o de interIeuquina- 2, a lo largo de cuatro semanas, sobre el tumor Il-7 implantado en el tejido subcutáneo epicraneal, no modifica el ritmo de crecimiento tumoral. Sin embargo, la administración intratumoral de células NK, células LAK y células TIL, consigue retrasar significativamente el ritmo de crecimiento tumoral. Estos hallazgos confirman resultados previos obtenidos en el caso de pacientes con gliomas malignos y plantean la conveniencia de nuevos estudios de inmunoterapia adoptiva con linfocitos activados en pacientes con tumores neuroectodérmicos indiferenciados.

Summary

An experimental study of adoptive immunotherapy on the experimental tumor IL-7 has been performed. This murine tumor shows features of primitive neuroectodermal neoplasm with capacity for neuroblastic differentiation. The results that we have obtained show that adoptive immunotherapy with cytokines such as interferon-alpha 2b or interIeukin-2 does not influence the growth rate of the IL-7 tumor, when it is located into subcutaneous epicranial space. Nevertheless, intratumoral administration of activated lymphocytes (NK, LAK or TIL-cells) achieves a clear delay in the growth rate of the IL-7 tumor. These findings agree with previous clinical observations about immunotherapy for malignant gliomas, and suggest the necesity of further studies in order to establish the usefulness of adoptive immunotherapy by means of activated lymphocytes in patients with undifferentiated neuroeetodermal neoplasms.
